technique for particle analysis

Dear,

I am a beginer in particle analysis to measure the pixel area and the number
of particles.
Enclosed is an example.
The island parts in white color should be treated as an indepent particle.
But my ImageJ figured out only two small dots as particles.

Would you please give a comment how to manipulate it?

Best,

Lee

Dear Lee,

I could get ImageJ to see only the two "islands" if I first
used Image --> Lookup Tables --> Invert LUT to make the
islands black = 255 instead of white = 255.  I also checked
"exclude on edges" during the Analyze particles step.

Best,
Christine

Hi Lee,
To count white particles you first have to apply some form of
thresholding with the "dark background" option check. Also, if you want
to include the white "blob" at the edge of the image, then make sure
that the "Exclude on Edges" option is unchecked.

Gabriel
